The aim of the research is a comparative study of the reactions of spontaneous rosette formation for the determination of T-lymphocytes in the blood of healthy and tuberculosis-infected cattle. We used lymphocytes isolated from the blood and lymphoid organs of 72 healthy cows and 96 tuberculosis cows. The most effective method was the isolation of lymphocytes on 17% verografin. To prepare the suspension, medium 199, Igla, on phosphate buffer and in Hank’s solution were used, of which Hank’s balanced solution (pH=7.2) and phosphate buffer (pH=7.2) turned out to be the most suitable. It was determined that the majority of tuberculous lymphocytes adsorbed 1620 ram erythrocytes (35-45% of the total number of rosette-forming lymphocytes) on their surface. The greatest number of these cells was noted in the thymus of animals with tuberculosis. The relative content of rosette-forming lymphocytes in the peripheral blood of healthy cows was 48.85±3.27%. In individual animals, the population of these cells ranged from 24% to 72%. A sharper decrease (2.33±1.20%) of rosette-forming lymphocytes was observed in the group with pathologically anatomically pronounced tuberculosis. The absolute number of rosettes increased depending on the degree of development of the tuberculous process. Thus, it was found that the blood lymphocytes of healthy cows form spontaneous rosettes. The number of T-lymphocytes in the blood and lymphoid organs of sick cows was significantly reduced. Further studies are needed to finally explain the obtained contradictory facts.
