Molecular genetic analysis of bovine herpesvirus type 5 (BOHV-5) in the seminal fluid of breeding bulls in the farms of Vologda region

Summary. The common bovine viruses include a-herpesviruses, which have a wide range of natural hosts. For several years in the farms of Vologda region were observed mass diseases of calves with symptoms of respiratory pathology (cold, cough, fever). Cows and calves were diagnosed with vulvovaginitis. The bovine herpesvirus type 5 was found in samples of nasal discharge, faeces and milk. The results obtained earlier were the basis for the study of bull semen as a possible source of infection for the presence of the virus. Experimental studies were conducted in several farms of Vologda region, laboratory studies - in the laboratory of virology of the Federal Scientific Center - All-Russian Research Institute of Experimental Veterinary Science named after K. I. Skryabin and Ya. R. Kovalenko of the Russian Academy of Sciences (Moscow). Semen analysis using polymerase chain reaction technique revealed bovine herpesvirus genome fragments. As a result of the sequencing it is determined that this pathogen belongs to type 5. The resultant amplicons were studied by phylogenetic analysis, which showed that the fragments of the genome of new isolates are close to previously known sequences (AY 745876, P 160/96, ISO 97/45) in the GenBank database, and are a single genetic group. Studies have shown that infected bull semen is one of the main sources of the bovine herpesvirus of type 5 among dairy herds in the farms of Vologda region. Therefore, a targeted screening of the semen for bovine herpes infection type 5 is required, as well as close control of sperm production during the entire period of use of the bulls with 6-month-interval.
