Frequency of detection of bacteria of the pasteurellaceae family and influence of storage time of genetic material on pcr results

Summary. Pasteurella - gram-negative, facultative anaerobic rod-shaped bacteria of family Pasteurellaceae, are commensal inhabitants of the surface of the mucous membranes of the upper respiratory tract of animals that can cause secondary infectious diseases. Pasteurellosis urgency of the problem in recent years due to the intensification of livestock aimed at increasing of milk and meat productivity of cows, often stresses background, virus infections, and poor conditions of feeding and maintenance. It was found that in farms of 3 regions of Siberia there are two types of bacteria Pasteurella multocida A and D genotypes and Mannheimia haemolytica A1 in large horned cattle of different age and gender groups. The circulation of genotypes B, E and F is not set among the animals of examined farms. According to the multiplex PCR predominant genotype of Pasteurella multocida, circulating among cattle in dairy complexes in Siberia, was genotype A (43.5%). This pathogen gene was present in samples from up to 1 month calves (34.7%), calves at the age of 1-6 months (65.6%) and adult animals (6.6%). Genotype D revealed an average of 5.7% of the investigated samples. Two genotype Pasteurella multocida bacteria contained 5.9% of the samples. Mannheimia haemolytica revealed in average of 11.2% of cases. The most sensitive animal categories for this pathogen are heifers (25%), as well as calves at the age of 1-6 months (6,9-7 %% positive samples). It was established that the optimal temperature mode of the storage of animal internal organs' suspensions, grown bacterial colonies are selected DNA is minus 18°C for 6 months, regardless of the type of the material. PCR can effectively detect the genome of Pasteurella multocida and Mannheimia haemolytica bacteria in samples of biological material of different origin from diseased and infected animals.
