Diagnostic evaluation of PCR and RDIF using samples from field outbreaks of African swine fever

DIAGNOSTIC EVALUATION OF PCR AND RDIF USING SAMPLES FROM FIELD OUTBREAKS OF AFRICAN SWINE FEVER

Summary. Statistical parameters of diagnostic polymerase chain reaction and direct immunofluorescence reaction based on the use of complex project stratified sample test population of primarily infected domestic pigs and wild boars from field cases or outbreaks of African swine fever are presented in the article. The results of PCR testing of unpaired and paired samples tests of parallel evaluation of MsNemar’s test did not provide significant differences between PCR and RDF. This indicates chance of false-negative results of PCR with unpaired testing, i.e. the uncertainty caused by the effect of an empirical factor in project selection. In the study of paired samples from dead pigs were from ASF albeit minor, but the differences of sensitivity of PCR and RDIF were 2.64 ( 0,18-4,4 )% (P = 0.01), which is statistically even more pronounced when testing of random samplies of hunted boars - 6.99 ( 1,6-10,5 )% (P = 0.01). PPR values in the parallel system "PCR RDIF" were significantly higher for the confirmation of the ASF, from 95,7 (86,0-98,4) 10% in prevalence ( 3-24 % ) in large herds to 99.2 (98.5-99.5) and 99.6 % (99,3-99,7)% - in the medium and small (and 53 %, respectively). Almost parallel system PCR-RDIF has good reliability and reproducibility to confirm clinical cases of ASF in any herds in domestic pigs and wild boars without confirmation of positive results of the reference method.
